Genomic DNA Purification and Library Preparation for Readout Sequencing
Genomic DNA from myoblasts and myotubes was purified using MasterPure™ DNA Purification
Kit (MC85200) according to manufacturer’s instructions. We performed two successive PCR
reactions (18 cycles for the first and 12 cycles for the second reaction) to add barcodes, stagger
and Illumina sequencing primers using Herculase II Fusion DNA Polymerase (Agilent, #600675)
(12). PCR products were purified by Agencourt AMPure XP magnetic beads and the DNA was
analyzed by Qubit Fluorometric Quantitation. The purified PCR product was sequenced on a Hiseq 2500 with 75 bp single reads.
