Resuspend the cell pellet in 200 μL of Muse™ Cell Cycle Reagent.

Direct cell counting was performed after Nigrosin Blue (0.1% (w/v)) staining using a haemocytometer and light
microscopy; cells which excluded the dye were considered to be viable. MTS assays were performed according to the manufacturer’s instructions. For clonogenic assays, the colonies formed following 3 weeks of culture were counted after staining withCrystal Violet (0.5% (w/v) in methanol).Cell cycle analysis was performed on a Muse Cell Analyzer using a Muse Cell Cycle Assay Kit according to the supplied instructions."
