PI/RNase Staining Buffer

Cell cycle assay human - U87

Experiment
Cell cycle assay human - U87
Product
PI/RNase Staining Buffer from BD Biosciences
Manufacturer
BD Biosciences

Protocol tips

Protocol tips
- Cells were fixed for at least 2 h, and stored at −20 °C until analysis
Downstream tips
- Store tubes at 4°C protected from light prior to analyzing.

- Analyze on the flow cytometer within 1hour

Publication protocol

Cell cycle was analyzed by flow cytometry (BD FACS verse Instrument) using the data acquisition BD FACsuite
software. Briefly, subsequent to transfections, cells
were trypsinized, collected into eppendorf tubes, washed with PBS by centrifugation at 700g, and fixed in 70 % ethanol at −20 °C for at least 2 h, and stored at −20 °C until analysis. Cells were then washed again with PBS and stained with Propidium Iodide (PI)/RNase Staining Buffer solution (BD Pharmingen) at room temperature in the dark, and PI fluorescence was analyzed by flow cytometry (protocol as described by BD Biosciences).
