Protocol tips

Upstream tips
Seed 1.5 × 105 cells/well in 6-well plate.
Protocol tips
Fix cells with Fixative Solution for 15 min at room temperature.

Add 1 ml of the β-Galactosidase Staining Solution and incubate the plate at 37°C at least overnight in a dry incubator (no CO2)

Publication protocol

SA-β-gal staining was performed using the Senescence β-Galactosidase Staining Kit (No. 9860S, Cell Signaling Technology) according to the manufacturer's instructions. Briefly, hMSCs were seeded at a density of 1.5 × 105 cells/well in 6-well plates. After the corresponding treatments, hMSC were washed twice with PBS and fixed with Fixative Solution for 15 min at room temperature. After fixation, the hMSCs were stained with 1 mL complete β-gal staining solution per well overnight at 37 °C. The complete β-gal staining solution contained Staining Solution, Solution A, Solution B and X-gal solution (final concentration of 1 mg/ml). After staining, the cell images were captured using a microscope (IX73, Olympus).
