Fix cells for 5 minutes in 3.7% formaldehyde..

Add 1 ml of the β-Galactosidase Staining Solution and incubate the plate at 37°C at least overnight in a dry incubator (no CO2)

Publication protocol

Senescence-associated β-gal staining was performed using the senescence-galactosidase staining kit (Cell Signaling Technology) according to the manufacturer's instructions. Briefly, cells were fixed for 5 minutes in 3.7% formaldehyde, washed with PBS and stained in β-galactosidase staining solution at 37°C until the intensity of staining was visible in either experimental or control plates. The number of positive cells was counted under the light microscope.

For tissue staining, mice were killed using carbon dioxide followed by isolation of the hearts. The heart tissues of wild-type and transgenic mice were excised, rapidly frozen in liquid nitrogen and mounted in OCT. Sections (4 µm) were cut, mounted onto glass slides, fixed in 2% formalin in PBS for 1 minute at room temperature, and immersed in β-gal staining solution at 37°C for 24 hours. The samples were counterstained with eosin and viewed under brightfield at 40× magnification.
