Frozen yeast/bacterial samples should be thawed and equilibrated to room temperature (15–25°C) before beginning the procedure.

Publication protocol

Bacterial genomic DNA was extracted from 1·5 mL broth cultures by the use of a Puregene DNA isolation kit (Gentra System‐Flowgen) following the manufacturer's instructions; the DNA eluate was resuspended in 150–200 μL elution buffer. DNA was extracted from 500 to 1000 μL crude plant extract (samples with symptoms) and concentrated plant extract (symptomless sample) using a DNeasy Plant Mini Kit (QIAGEN), according to the manufacturer's instructions. DNA concentrations were determined using a NanoDrop ND‐1000 spectrophotometer (Thermo Fisher Scientific), then diluted in sterile milli‐Q water to a final concentration of 5 ng μL−1 and stored in aliquots at −20°C.
