RNA isolation / purification Tissue - Human Muscles

For RNA extraction, the Maxwell 16 LEV simplyRNA Tissue Kit was used (Promega), following the manual's instructions. Briefly, minced and ground human muscle tissue was incubated in 200 µl of chilled 1-Thioglycerol/Homogenization solution and further homogenized using the Precellys 24 system (see DNA). Afterwards, the samples were heated at 70°C for 2 min, then the lysates were allowed to cool down. 200 µl of lysis buffer was added to the cooled-down homogenate, mixed vigorously, followed by transfer of the total 400 µl into well 1 of the Maxwell 16 LEV cartridge. 5 µl of DNAse was added to well 4 of the cartridge and, 50 µl RNAse-free water was added to 0.5 ml Elution Tubes and the RNA extraction program was started at the Maxwell 16 instrument.Full paper Login or join for free to view the full paper.

RNA isolation from tissue
How do I extract RNA from animal tissue without using liquid nitrogen? I tried the RNA extraction by using the TRIzol reagent and I homogenize the tissue using polytron homogenizer at room temperature for 30secs is this correct?

Problem in phase separation after using serum/plasma kit
I used a serum/plasma kit for my serum samples. After the phase separation the samples should have 3 phases: a colourless aqueous phase, a white interphase and a red organic phase. However, in some of my samples there was no aqueous phase unless I wait for an extended period of time. How can I circumvent this problem?
