Do not include serum and antibiotics during the formation of the ICAFectin 442/siRNA complexes.

Publication protocol

Transient transfections of HeLa cells and COS-7 with siRNA and/or plasmids were performed using ICAFectin442 (Eurogentec), JetPrime (Polyplus), or FuGENE 6 (Roche) following the manufacturer's instructions. HEK293T cells were transfected with the calcium phosphate transfection method.
Stable transfection of HeLa cells with shRNA or plasmids was performed using FuGENE 6. HeLa clones were obtained by limiting dilutions. For generation of retroviral supernatants, pMSCV-puro vector (encoding different Optn constructs or control vector) was transfected into Plat-A packaging cell line using FuGENE 6. High titers of recombinant viruses were obtained 48 hr after transfection and used to infect Optn-depleted HeLa cells.
