26. We use cationic lipid-based transfection reagents (Felgner and Ringold, 1989), Lipofectamine® 2000, Lipofectamine® 3000, and non-liposomal FuGENE® HD formulation to transiently transfect the 293FT cells with 2.5 μg of AtGORK plasmid obtained in step 10 according to the respective manufacturer's instructions (Table ​(Table1).1). ♦ CRITICAL STEP Equilibrate the transfection reagents to room temperature before use and mix by inverting or through a brief vortex. • OPTIONAL We recommend the use of OPTI-MEM® I (1X) Reduced Serum Medium as diluent in all transfection steps. Alternatively, serum-free DMEM (1X) + GlutaMAX™-I medium can be used for the transfection protocol. ❖ Troubleshooting (Table ​(Table22).
