Briefly, cells were cultured in the presence or absence of lysosomal proteases for 2 h following which, cells were collected and lysed and subjected to immunoblotting for LC3-II or p62.

Publication protocol

Cell extracts were prepared from 293T cells that were transfected with HA-Beclin 1, Myc-Rac1, Myc-Rab5, and Myc-Cdc42 and incubated with an anti-HA antibody for 2 h at 4 °C. Immunoprecipitates were prepared by incubation with Dynabeads protein A (Invitrogen) and subjected to immunoblot with anti-Myc antibody.
