The KU812 cell line" was maintained in RPMI-1640 medium containing 10% fetal calf serum (FCS), 2 mM L-glutamine and antibiotics (100 pg/ml gentamycin) (Flow Laboratories, Irvine, U.K.). The cells were passaged every 3-4 days. To test for basophilic differentiation, KU812 cells were cultured at 3 x 105 cells/well in 96-well plates. Each well contained 250 yl of medium supplemented with 0-3 mm sodium butyrate (Sigma,
St Louis, MO) as described previously. 12 After 3 days of culture the cells were resuspended and samples were harvested for analysis. Cell growth was determined by counting of viable cells with the use of a Buerker haemocytometer and trypan blue exclusion.
