Protocol tips

Publication protocol

After cell fixation, BrdU was incorporated into DNA ends using terminal deoxynucleotidyl transferase (TdT)(Phoenix Flow Systems, AU: 1001). The reaction was performed at 370C for 1 hour. Subsequently, the samples were incubated for up to 12 hours in 1% BSA and then BrdU was recognized by a primary anti-BrdU mouse monoclonal antibody (Abcam, ab8039). Fluorescent signal was generated by incubation with a secondary goat anti-mouse antibody conjugated with Alexa Fluor 488 (Invitrogen, Thermo Fisher Scientific, A-11029).
