Apoptosis assay cell type - CaOV-3 human ovarian cancer

A gross majority of classical apoptotic attributes can be quantitatively examined by flow cytometry, the preferred platform for rapid assessment of multiple cellular attributes at a single-cell level. However, sample preparation for such flow cytometry-based techniques could be challenging. Cell harvesting by trypsinization, mechanical or enzymatic cell disaggregation from tissues, extensive centrifugation steps, may all lead to preferential loss of apoptotic cells. To overcome this strictly follow manufacturers instruction of the detection kit.

Upstream tips
- 10,000 cells per well were platted for an assay

- Cells were pre treted with ABC294640, paclitaxel, or vehicle for 24 h.
Downstream tips
- When the absorbance of
samples too high, reduce agent-concentration and decrease incubation period.

- When the absorbance of
samples too low, increase the agent concentration and prolong the agent incubation interval
Upstream tips
- Cells were assayed after they were transfected with miR-340 mimic for 24 h.
Protocol tips
- Treated with FITC Annexin V and PI and incubated for 15 min at RT (25°C) in the dark
Protocol tips
Add 5 μl of Annexin V-fluorescein isothiocyanate and 5 μl of 7-AAD.
