Quantitative RT-PCR and mtDNA content analyses.
Total RNA was extracted using REzol (PROtech, Mukilteo, WA). Samples of mRNA were analyzed by SYBR green-based real-time quantitative reverse transcription (RT)-PCR (Applied Biosystems, Foster City, CA) with the gene for cyclophilin A as the reference in each reaction. Total DNA was extracted from cells using a genomic DNA isolation kit (Geneaid, Taipei, Taiwan). The mtDNA content was calculated using real-time quantitative PCR by measuring a mitochondrial gene (Cox1) versus a nuclear gene (Gapdh).
