Live / Dead assay yeast - Urediniospore

Experiment
Live / Dead assay yeast - Urediniospore
Product
LIVE/DEAD™ FungaLight™ Yeast Viability Kit, for flow cytometry from Thermo Fisher Scientific
Manufacturer
Thermo Fisher Scientific

Protocol tips

Protocol tips
Add 1 µL of Component SYTO and 1 µL of Component propidium iodide.

After stain is added, each tube should be vortexed gently.

Incubate all samples at room temperature or 37ºC protected from light for 15–30 minutes

Publication protocol

Urediniospore viability was determined by three methods: (i) the FungaLight yeast vitality kit (Molecular Probes Inc., Eugene, OR) containing CFDA, acetoxymethyl ester (AM) and PI; (ii) the LIVE/DEAD yeast viability kit (Molecular Probes Inc.) containing FUN 1 and calcofluor white M2R; and (iii) an in vitro germination test. Three spore samples (live, heat-killed, and a 1:1 mix) were tested for each method.
