Protocol tips
- 5 x 106 cells/mL can be seeded for an assay and fixed with 1% paraformaldehyde in PBS, pH 7.4, for 10 minutes

- 3.0% hydrogen peroxide was used for quenching.

Publication protocol

TdT-mediated dUTP nick end labeling (TUNEL) was performed with an ApopTag Plus Peroxidase In Situ Apoptosis Detection Kit (Merck Millipore, Darmstadt, Germany) according to the manufacturer’s instructions. The number of TUNEL-positive cells was counted and averaged across 10 random fields from 7 mice per group.

Are 10 week old samples in formalin still usable?

I have left my tumor samples in formalin for an extended period of time (around 10 weeks). Do you think I will be able to use them for TUNEL assay and get results? Thank you for your help in advance
