Human lung adenocarcinoma A549 (CCL-185, ATCC) cells were cultured in Dulbecco's modified Eagle's medium or RPMI 1640 medium (Invitrogen) supplemented with 10% heat-inactivated fetal bovine serum (Invitrogen). They were maintained at 37 °C in 5% CO2. Nocodazole, cyclohexamide (Sigma), MG132, U0126, LY294002 (Cayman Chemical Co., Ann Arbor, MI), SB203580, PD98059, and SP600125 (Cayman Chemical Co.) were dissolved in DMSO. HtrA2 (catalogue no. 35615) was silenced using a commercial siRNA kit (Santa Cruz Biotechnology, Inc.). A nonspecific scramble siRNA was the negative control. Transfection was performed by electroporation using a pipette-type microporator (Microporator system, Digital BioTechnology, Suwon, Korea). |
