Publication protocol
Total intracellular ROS and mitochondrial ROS levels were measured by DCFH-DA and MitoSOX, respectively. Human prostate cancer PC-3 cells were cultured in a 6-well plates at a density of 5×104 cells/well. After exposure to DPT for the indicated times, the cells were incubated with DCFH-DA or MitoSOX at a final concentration of 10 μM or 5 μM for 15 min at 37°C, respectively. Cells were washed twice with PBS and fluorescence positive cells were measured with flow cytometry and analyzed with CellQuest analysis software.
