Publication protocol
The BD Biocoat invasion kit (BD) was used to evaluate the tumor invasive ability according to the manufacturer's protocol. Briefly, around 5×104 cells of PANC-1 and BxPC-3 with basal media were transferred in each 6-well upper chamber in the presence or absence of OPP. In the meantime, 3 mm of culture medium with 10% FBS was added into each lower chamber of a 24-well plate. After 20 h of incubation, the cells in the upper chamber were removed using a cotton stick. Later, the cells that invaded through the Matrigel matrix membrane were stained with 4 μg/ml Calcein AM in Hanks buffered saline at 37°C for 1 h. Each of the experimental conditions was performed in triplicates. Subsequently, fluorescence of the invaded cells was read in an ULTRA Multifunctional Microplate Reader (Tecan) at excitation/emission wavelengths of 530/590 nm. These fluorescently labeled invasive cells were also photographed under a fluorescent microscope.
