Migration assays were performed in 6.5 mm diameter transwell plates containing 8 μm pore size filters (Corning, Costar). Briefly, transwell analysis was conducted after cells were harvested and resuspended (2.0 × 10 cells/ml) in serum-free growth medium. 100 μl of cell suspension (control and treated) was added to the upper chamber whereas in the lower chamber 750 μl of growth medium with 10% fetal calf serum was added with or without SP and LPS. The control group was induced by 10% FCS alone. After 24 h incubation at 37°C, the transwells were removed, the cells were fixed with 4% paraformaldehyde, permeabilized with methanol 100%, colored with crystal violet and then the cells on the upper side were scraped off. The number of cells was counted in 5 random fields under 40 × magnification, and the mean was calculated. The experiments were repeated three times
