- The pretreated cells plus RA SF (1:20) or flagellin (10 ng/ml) were added to the polymerized matrigel and the plate was incubated for 16h at 37°C

Publication protocol

To perform the matrigel tube formation assay, BD Matrigel Matrix (BD BioCoat tube formation kit, 50 µl) was polymerized for 30 min at 37°C in a 96-well plate. In order to determine whether TLR5 ligation is pivotal for RA SF induced endothelial tube formation, HMVECs/HUVECs were incubated with antibody to TLR5 or IgG (10µg/ml) for 45 minutes at 37°C. The pretreated cells plus RA SF (1:20) or flagellin (10 ng/ml) were added to the polymerized matrigel and the plate was incubated for 16h at 37°C. Each condition was performed in triplicate, bFGF (20 ng/ml; R&D systems) was used as a positive control, and PBS was used as a negative control. Following incubation, the number of branch points/tubes was quantified as previously described (17, 19, 20).
