Add 10 μl of the MTT
labeling reagent (final concentration 0.5 mg/ml) and incubate the microplate for 4 h in a humidified atmosphere (e.g., +37°C, 5 - 6.5% CO2 )

Publication protocol

The Cell Proliferation Reagent Kit I (MTT; Roche Applied Science) was used to test the cell viability. A549 and PC-9 cells plated in 96-well plates (3,000 cells/well) were transfected with si-PVT1. We estimated the cell viability every 24 hours according to the manufacturer's protocol. The MTT experiments were conducted in quadruplicate. For colony formation assay, transfected cells (n = 1200) were placed in 6-well plates and maintained in proper medium containing 10% FBS for 2 weeks. The medium was replaced every 4 days, and visible colonies were counted after 2 weeks.
