Publication protocol

HUVECs (2×105) in 6-well plates were transfected with NFκB promoter-luciferase (Clontech, CA, USA) and β-galactosidase plasmid for 48 h using lipofectamine 2000 according to the manufacturer’s instructions. After co-incubation with dexmedetomidine and LPS for 48 h, cell lysates were assayed for luciferase activity and β galactosidase activity using a Secrete-PairTMDual Luminescence Assay kit (Gene Copoeia, MD) as measured with a microplate reader (SpectraMax MAX190 spectrophotometry).
