Publication protocol

Senescence-associated β-galactosidase (SA-β-gal) staining was performed using a β-galactosidase Staining Kit (Beyotime, Shanghai, China) according to the manufacturer’s instructions. Briefly, 5 × 10 hASCs (passage 5) were seeded onto 6-well plates and incubated with freshly prepared β-gal staining solution overnight at 37 °C in the absence of CO. hASCs were washed with PBS, and the blue color (i.e., senescent cells) was observed under microscopy. Phase contrast images were taken, and the percentage of SA-β-gal-positive cells was calculated by dividing the number of blue cells by the total number of cells, multiplied by 100.
