Senescence-associated β-galactosidase staining (CST, USA) was performed according to the manufacturer's protocol. Briefly, cells in a 6-well plate were washed with PBS, fixed for 15 min at room temperature with fixative solution, and incubated for 24 hours with β-galactosidase staining solution in a dry incubator (absence of CO2). H9c2 cells were observed for the development of blue color under a microscope (100x). The percentage of positive cells was calculated by counting the blue-stained cells and total cells (as a standard) in five randomized fields.
