2.4. Genome-wide methylation assay
The genome-wide methylation assay was carried out as described previously [23]. Briefly, 2 μg of chromosomal DNA isolated from a 60-mm culture dish was applied to an Illumina Infinium Human Methylation EPIC Beadchip (Illumina, San Diego, CA, USA) to detect genome-wide methylation. A methylation index (β) was calculated for each site, which is a variable ranging from 0 (no methylation) to 1 (100% methylation). The β value was then used to calculate a ratio of relative methylation between the control and Rh2-treated cells. The array data is accessible from the Gene Expression Omnibus database (http://www.ncbi.nlm.nih.gov/geo/) with the accession number GSE93356.
