The conventional PCR amplification of
the D-loop region was carried out in a 25 µl
reaction volume consisting of 1 U Platinum Taq
DNA polymerase (Invitrogen), 5 M each D-loop
primer (Applied Biosystems), 4x reaction buffer,
containing dNTPs (Invitrogen) and 25 ng of total
DNA. The thermocycling conditions included an
initial denaturation phase at 94°C for 2 min, then 35
cycles of denaturation at 94°C for 15 s, annealing
at 60°C for 30 s, and extension at 72°C for 1 min,
followed by a final extension at 72°C for 5 min
