1.5 × 105 SUIT2 or PANC-1 cells were seeded in 6-well plates and allowed to adhere overnight before adding 300μl transfection medium (Santa-Cruz) containing 3μl CRISPR Transfection Reagent (Santa-Cruz) and 2μg of control (Santa Cruz - sc-418922) or DCK CRISPR plasmid (Santa-Cruz - sc-417715). 24 hours following transfection, GFP positive cells were isolated by flow cytometry and cultured in DMEM media containing 20% FCS. Following recovery, cells were maintained under normal culture conditions and protein and cell viability assays performed as described above.

DNA insert using CRISPR

I would like to excise a large strand of DNA and insert a new one using CRISPR. My problem is that my strand will be a little over 1kb and I am not sure if this is going to be a limiting factor. Also, how long should the homology arms be for a region of this size?
