Cell migration was evaluated by the wound healing assay using the CytoSelect Wound Healing Assay Kit (Cell Biolabs) as described previously.16 Briefly, wound healing inserts were put into 24-well cell culture plates. C166 cells (250 μL) transduced with Ad-GFP or Ad-shRGC32 were added to either side of the insert and incubated overnight to form a monolayer. Then the inserts were removed, and the cells treated with vehicle or VEGF (20 ng/mL) for 24 h to allow the cells to migrate. Images of wound healing were captured using a dissection microscope at a magnification of ×40. Cell migration was quantified by blindly measuring the migration distance.
