Seed 0.5×10^6 cells/well

siRNA concentration-50 nM.

Incubate HiPerFect Transfection Reagent to diluted siRNA for 10–15 min at RT.

Incubate the cells with the transfection complexes under their normal growth conditions for 96h.

Publication protocol

BMDM were detached from the dish by incubation in PBS/5 mM EDTA and seeded at 0.5×106 cells/well in a 6-well plate. The cells were immediately transfected with 50 nM siRNA pool or individual siRNA targeting mouse SAMHD1 (Dharmacon) using HiPerfect lipofection reagent (Qiagen). The efficiency of knock-down was assessed 4 days post-transfection by immunoblot analysis.
