Publication protocol

Human and mouse COX‐2 siRNAs were purchased from Santa Cruz Biotechnology. Each siRNA (mixture of 50 nM COX‐2 and 50 nM control siRNA) was transfected into cells using Lipofectamine reagent according to the manufacturer's protocol. The transfected cells were cultured in DMEM. WM‐115 cells were cultured in Eagle's minimum essential medium without antibiotics for 8 h, cultured in complete medium for 36 h, harvested, and subjected to the cell migration assay. Knockdown of COX‐2 expression in cells after transfection was confirmed using Western blot analysis.
