Publication protocol

Protein levels for candidate genes were determined by quantitative western blot. Each experiment was assessed on a single western blot to ensure identical analysis conditions between treated and control animals. Total protein was isolated from mouse liver using Tissue Extraction Reagent I (Life Technologies, Burlington, ON) supplemented with cOmplete protease inhibitor cocktail (Roche, Laval, QC). Protein extract, diluted 1/10 and 1/20 with 1XPBS, was quantified by Bradford assay and diluted to a final concentration of 10 µg/µL. A total of 65 µg protein, was loaded into each well of a Novex 4–12% Bis-Tris midi-gel system to ensure sufficient material would be available for the detection of low abundance targets.
