Publication protocol
The pyruvate decarboxylase activity of strains No. 2201 and KY2132 was measured as described in Flikweert et al. (1996). Cell extracts were prepared from yeast cells, incubated in 5 ml YPD10 to late log phase at 30 °C, using CelLytic Y Cell Lysis Reagent (Sigma). Pyruvate decarboxylase activity was assayed at 30 °C immediately after extract preparation, using a Molecular Devices VersaMax microplate reader set at 340 nm. Protein concentrations in cell‐free extracts were determined using Protein Assay CBB Solution (Nacalai Tesque) with bovine serum albumin (Sigma) as a standard.
