HT1080 cells were transfected with 10 µm 2–5A or PolyI:C (2 µg/mL) using Lipofectamine 2000. After 6 h total RNA was isolated from transfected cells using Trizol reagent (Invitrogen) and quantitated by measuring absorbance at 260 nm. RNA (2 µg) was separated on RNA chips and analyzed with Bioanalyzer 2100 (Agilent Technologies, Santa Clara, CA, USA) as described previously [48]. Total RNA was isolated from HT1080 cells transfected for 6 h with 2–5A or mock-transfected with transfection reagent alone using Trizol Reagent (Life Technologies, Carlsbad, CA, USA). Control RNAs were purified from cells that were not transfected with 2–5A and would include other dsRNAs in cells not resulting from RNase L enzyme activity. Small (<200 nucleotides) RNAs were isolated using mirVana miRNA Isolation Kit (Ambion, Austin, TX, USA) as per manufacturer’s protocols and classified as RNase L-cleaved small RNAs or control small RNAs. RNA was quantitated using Nanodrop and complexed with Lipofectamine 2000 and introduced into cells.
