Publication protocol

Total RNA was isolated from LSCs using the High Pure RNA isolation kit (Roche Diagnostics GmbH, Germany) according to the manufacturer's instructions. BCR/ABL translocation and Telomerase mRNA levels in LSCs were measured by using a qRT-PCR method. Primers and probes were designed with the Universal Probe Library Assay Design Center (https://www.universalprobelibrary.com). For each reaction the complementary DNA (cDNA) equivalent was amplified for 35 cycles using cycle parameters of 94°C for 30 seconds, 60°C for 60 seconds, and 72°C for 60 seconds. All PCR reactions were performed in the LightCycler® 480 instrument (Roche Diagnostics, Germany). Glyceraldehyde 3-phosphate dehydrogenase was used as the housekeeping gene in order to normalize expression levels. Each sample was tested in triplicate.
