- 1-Bromo-3-chloropropane is less toxic than chloroform and its use for phase separation decreases the possibility of contaminating RNA with DNA

- The chloroform used for phase separation should not contain isoamyl alcohol or other additives.

GFP+ cells from lactating Adipoq-Cre; mT/mG mice were sorted directly into Trizol LS (Invitrogen, 10296-028); total MG tissue from GW9662-treated and Adipoq-Cre; mT/mG; iDTR (and associated control) mice was flash frozen and subsequently thawed and resuspended in Trizol (Sigma, T9424). RNA extraction and purification was performed using the RNeasy mini kit (QIAGEN 74104) following manufacturer instructions. cDNA was generated using equal amounts of total RNA with the Superscript III First-Strand Synthesis System (Invitrogen 18080051) using Oligo dT per the manufacturer’s instructions. Real-time PCR was performed as previously described16,19 using SYBR green I Master mix (Roche 04887352001) on a LightCycler 480 (Roche)

RNA isolation from tissue

How do I extract RNA from animal tissue without using liquid nitrogen? I tried the RNA extraction by using the TRIzol reagent and I homogenize the tissue using polytron homogenizer at room temperature for 30secs is this correct?

Problem in phase separation after using serum/plasma kit

I used a serum/plasma kit for my serum samples. After the phase separation the samples should have 3 phases: a colourless aqueous phase, a white interphase and a red organic phase. However, in some of my samples there was no aqueous phase unless I wait for an extended period of time. How can I circumvent this problem?
