- For low 260/230 readings the best approach is to try washing sample (precipitation) with ethanol to desalt it.

Publication protocol

To determine the mRNA expression of ICAM-1, the total RNA was isolated from cells using TRIzol reagent (Invitrogen; Thermo Fisher Scientific, Inc., Waltham, MA, USA), according to the manufacturer's protocol. RNA was frozen at −80°C until analyses were performed. Any DNA contamination in the isolated RNA was eliminated with DNase treatment. The RNA concentration was determined spectrophotometrically at 260 nm (SmartSpec™ 3000; Bio-Rad Laboratories, Inc., Hercules, CA, USA). The RNA integrity was evaluated by 1% agarose gel electrophoresis. Of the total RNA, 3 µg was used for the RT reaction using the ThermoScript RT-PCR systems (Invitrogen; Thermo Fisher Scientific, Inc.), according to the manufacturer's protocols.
