The following might help to increase RNA yield
- Heat the DEPC Water to 70°C before adding to the column.
- Increase the incubation time to 5 minutes.
- Increase the elution volume.
- Repeat the elution step with fresh DEPC Water (this may increase the yield, but decrease the concentration).
- Repeat the elution step using the eluate from the first elution (this may increase yield while maintaining elution volume). |

Publication protocol
RNA was isolated using an E.Z.N.A.® Yeast RNA Kit (OMEGA). Isolated RNA was treated with RNase-free DNase I (Fermentas) to remove contaminating genomic DNA. RNAs were reverse-transcribed using the iScript cDNA Synthesis Kit (Bio-Rad). Quantitative Real-Time RT-PCR (qRT-PCR) was performed using SYBR Select Master Mix (Life Technologies) with each primer sets (Supplementary Table S2). All reactions were performed in triplicate. Data analysis was performed by StepOneTM software. The threshold cycle number (CT) values were normalized by subtracting the mean CT value of histone H2B (htb1) mRNA. The relative levels of expression of mitochondrial-encoded RNAs (mt-RNAs) were calculated by using the 2−ΔΔCt method.
