Publication protocol

Expression of IκBα was selectively silenced utilizing predesigned siRNAs (Cell signaling). Scrambled siRNA was used as a non-targeting control. Caco-2 or HT-29 cells were transiently transfected with 100 pmol of siRNA duplexes for 48 h using Lipofectamine 2000 (Invitrogen, Grand Island, NY). Silencing was validated by real-time PCR and Western blotting utilizing IκBα specific primers and specific antibodies (Abcam, Cambridge, MA)
