Publication protocol

The detection of apoptotic nuclei using the TUNEL staining was performed with ApoAlert™ DNA Fragmentation Assay Kit (Clonetech, Palo Alto, CA). Briefly, cells were fixed in 4% paraformaldehyde and incubated in 0.3% H2O2 in methanol for five minutes to block endogenous peroxidase. The slides were then incubated with fluorescent-conjugated avidin in PBS for two hours at room temperature. After three washes, slides were cover slipped with glycerol phosphate buffer. Apoptotic cells were counted in at least 10 random fields and expressed as a percentage of the total cell number (apoptotic index).
