MRC5 cells (106) were cultured in in a 25-ml culture flask for 24 h, then treated with 4 µM PAB and/or 2 mM 3MA for 36 h. Adherent and floating cells were collected and frozen at −80°C. Western blot analysis was performed as described in previous reports (5,20–23). Protein expression was detected using the aforementioned primary antibodies (dilution, 1:1,000) followed by a corresponding AP-conjugated secondary antibody (dilution, 1:1,000). Proteins were visualized using nitro-blue tetrazolium and 5-bromo-4-chloro-3-indolyl phosphate.
