NHFs and U20S (expressing inducible p85β) were cultured in serum-free medium for 48–72 h to monitor the autophagic response. Recovery from the autophagic response was studied by replacing the serum-free medium with medium containing 10% fetal bovine serum for up to 3 h. Cell lysates were the subjected to SDS–PAGE and western blotting using antibodies against LC3-II and p62. U2OS cells stably expressing GFP–LC3-II were grown in serum-free medium for 1–3 h to monitor GFP–LC3-II using an inverted microscope, as described below.
