INS-1E cells grown on glass coverslips were fixed for 5 min in 4% ice-cold paraformaldehyde, rinsed in phosphate buffered saline (PBS) and permeabilized for 30 min in PBS supplemented with 1.5% BSA and 0.2% Triton X-100. Coverslips were incubated overnight at 4 °C in the presence of antibody against LC3B and p62 (dilution 1/100; see Table S2). Cells were then washed and further exposed for 1 h to an appropriate Alexa fluor 488-conjugated or 555-conjugated antibodies (1/1000; N.V. Life Technology AG, Switzerland), and photographed under fluorescence microscopy (Leica Camera AG, Nidau, Switzerland).
