Biopsies were collected in cold phosphate-buffered saline (PBS), cut into small pieces and digested with Liberase TM 0.56 U/ml (Roche, Basel, Switzerland) and DNAse 0.002% (Sigma, St. Louis, MO, USA) in DMEM:F12 (Invitrogen, Carlsbad, CA, USA) at 37 °C for approximately 4.5 h with magnetic stirring. Then, the samples were washed once and further incubated for 10 min in Trypsin–EDTA 0.05% (Invitrogen). The cell suspension was washed once with DMEM:F12 and seeded into 6-well plates at a concentration of approximately 1 mg tissue/cm2 in medium consisting of DMEM:F12 supplemented with 10% human serum, penicillin/streptomycin (PEST, PAA Laboratories, Pasching, Austria) and L-glutamine (Invitrogen). The cells were cultured for 10 days.
