Human chondrocytes were isolated from patients undergoing total knee arthroplasty. Non-OA knee samples were provided by National Disease Research Interchange (Philadelphia, PA, USA). The average age was 62 yr (range, 52–71). Chondrocyte isolation and culture were performed. Monolayer cultures were maintained in DMEM supplemented with 10% fetal bovine serum (FBS), 50 U/ml penicillin, and 50 μg/ml streptomycin (Biologic Industries, Beit-Haemek Kibutz, Israel). The cells were treated with the proinflammatory cytokines IL1β (5 ng/ml) and TNF-α (50 ng/ml; both from Peprotec, Rehovot, Israel) in defined serum-free medium (BIO-MPM 1; Biologic Industries, Beit-Haemek Kibutz, Israel). Conditioned medium was filtered with a 10-kDa cutoff Amicon filter (EMD-Millipore, Billerica, MA, USA) and processed for immunoblot analysis.
