Primary human coronary artery endothelial cells (HCAEC; purchased from Cell Applications, San Diego, CA, USA) were cultured (37°C, 5% CO2) in MesoEndo medium (Cell Applications). Cell viability was determined by lactate dehydrogenase (LDH) release [16]. Cells were passaged at ∼80% confluence, and used from passages 4–8. Cells were detached using trypsin-EDTA (Thermo Fisher Scientific, Scoresby, Victoria, Australia). For intact cell experiments, cells were diluted with 10 mL medium to inactivate trypsin, pelleted (5 min, 930×g, 21°C) and resuspended at 1–1.5×105 cells/mL. Aliquots (2 mL) were transferred to 6-well plates and incubated overnight before use. For lysate studies, trypsinised cells were washed twice in PBS (Astral Scientific, Caringbah, NSW, Australia), and then lysed in water (2×107 cells/mL) prior to freezing at −80°C.
