- Astrocytes are harvested and lysed follwoing the Mammalian Cell Lysis kit manufacturer's protocol.

Astrocytes were harvested and lysed using the Mammalian Cell Lysis kit (Sigma-Aldrich, MCL1-1KT). Lysates were centrifuged at 12000 × g for 10 min at 4°C and the protein content of the supernatant fraction was quantified by a BCA assay using the Pierce™ BCA Protein Assay Kit (Thermo Fisher Scientific, 23227) according to the manufacturer's guidelines. Equal amounts of soluble proteins were loaded and electrophoresed in a 10% or 15% sodium dodecyl sulfate-polyacrylamide gel under reducing conditions, followed by blotting onto a polyvinylidene fluoride membrane (Millipore, IPVH00010).
