Cells were lysed in Laemmli sample buffer and boiled at 96°C for 5 min. Cell lysates were sonicated by probe sonicator to sheer cellular debris and genomic DNA. To detect tubulins, lysates were diluted 1:10 in sample buffer. Proteins were separated by 10% Bis-Tris gel (Invitrogen) and transferred to nitrocellulose membrane. After blocking in 5% milk/TBS or BSA/TBS, membranes were incubated with primary antibodies at 4°C overnight and 1 h with secondary antibodies. Image acquisition was performed with an Odyssey imaging system (LI-COR Biosciences) and analyzed with Odyssey software.
