Publication protocol

Cells were transfected with siRNAs using Lipofectamine RNAiMax according to manufacturer's protocol. siRNAs targeted: human SLC16A1/MCT1 5′-AAG AGG CUG ACU UUU CCA AAU-3′ (siMCT1), human HIF-1α 5′-AAC UGG ACA CAG UGU GUU UGA-3′ (siHIF-1α), human EPAS1/HIF-2α 5′-CCC GGA UAG ACU UAU UGC CAA-3′ (siHIF-2α), and human c-Myc 5′-GAG AAC AGU UGA AAC ACA A-3′ (siMyc-1) and 5′-GAG AAC AGU UGA AAC ACA A-3′ (siMyc-2). ON-TARGETplus smartpool siRNAs (5′-CCC AAG AAC UAU ACU GAU A-3′, 5′-GCG AAG CGC UGU UGC UGU C-3′, 5′-GAA GAU CUA UGG UUG ACU A-3′, 5′-CCC AUG AAG UGC UAG AUA A-3′; Dharmacon) were used to silence human GLUD1/GDH1. Mock transfection and Allstar siRNA (Qiagen) were used as negative controls.
