Publication protocol

in vitro deubiquitination assay, beads were resuspended in 1× Deubiquitinase buffer (125 mM HEPES ([pH 7.5], 25 mM MgCl2, 10 mM NaF, 10 nM okadaic acid, 500 mM NaCl, 2.5 mM DTT, 0.5 mg/ml BSA). 10 µl of recombinant His-MYSM1 (1 µg) was then added to the bead slurry, followed by incubation for 30 min at 37 °C with gentle shaking. The reaction was then stopped by adding 2x SDS loading buffer followed by western blot analysis for anti K63, K48, K27, and M1 ubiquitin antibody.
